Abstract

The research aims at identifying amylase enzyme activities in human urine, determining the amounts of the activities with pure a-amylase enzyme as a comparison, and determining whether there was any difference between urine amylase enzyme and pure a-amylase enzyme. The research subject was potato starch and the objects were human urine amylase enzyme activity and pure a-amylase enzyme described as glucose levels of the hydrolysis of potato starch. Determination of amylase enzyme activities was done through two steps. The first step was a qualitative test of the amylase enzyme using Wohlgemuth method and the qualitative glucose test using Molish test and Benedict test. The second step was determining the optimum condition of the urine amylase enzyme activities and a-amylase pure enzyme activities involving determining the incubation time, pH, and the optimal temperature and determining the activities on substrate concentration variations of 0.25 % (w /v), 0.50 % (w /v), 0.75 % (w /v), 1 % (w /v), 1.50 % (w /v), and 2 % (w /v). The results showed that there were human urine amylase enzyme activities on potato starch using pure a-amylase enzyme as a comparison. The amounts of pure a-amylase enzyme activities on the variation of substrate concentrations of 0.25 %, 0.50 %, 0.75 %, 1 %, 1.50 %, and 2 % (w/v) amounted to 1.823 x 10', 1.883 x 10', 1.928 x 10', 1.98 x 10', 2.25 x 10', and 2.233 x 10' mg/ml.minute while the amounts of urine amylase enzyme activities were l.853 x 10', 2.148 x 10', 2.21 x 10', 2.493 x 10', 2.57 x 10', and 2.788 x 10' mg/ml.minute. The results also showed that there were differences in human urine amylase enzyme and pure a-amylase enzyme activities.
